The bioavailability of quetiapine is marginally affected by administration with food, with Cmax quetiapine seroquel: uses side effects and AUC values increased by 25% and 15%, respectively. Hypersensitivity to quetiapine or to any excipients in the SEROQUEL formulation. Anaphylactic reactions have been reported in patients treated with SEROQUEL. Seroquel tablets may be used as part of a treatment program to treat bipolar disorder and schizophrenia in children and teenagers. Seroquel extended release is used in combination with antidepressant medications to treat major depressive disorder in adults. Seroquel extended release is also used alone to treat manic episodes in children at least 10 years old with bipolar disorder. Quetiapine was administered in the diet to mice at doses of 20, 75, 250, and 750 mg/kg and to rats by gavage at doses of 25, 75, and 250 mg/kg for two years. These doses are equivalent to 0.1, 0.5, 1.5, and 4.5 times the MRHD of 800 mg/day based on mg/m2 body surface area (mice) or 0.3, 1, and 3 times the MRHD based on mg/m2 body surface area (rats). There were statistically significant increases in thyroid gland follicular adenomas in male mice at doses 1.5 and 4.5 times the MRHD based on mg/m2 body surface area and in male rats at a dose of 3 times the MRHD on mg/m2 body surface area. Mammary gland adenocarcinomas were statistically significantly increased in female rats at all doses tested (0.3, 1, and 3 times the MRHD based on mg/m2 body surface area). The development of cataracts was observed in association with quetiapine treatment in chronic dog studies see Nonclinical Toxicology. Lens changes have also been observed in adults, children, and adolescents during longterm SEROQUEL treatment, but a causal relationship to SEROQUEL use has not been established.
Elimination of quetiapine is mainly via hepatic metabolism with a mean terminal half-life of about 6 hours within the proposed clinical dose range. Steady-state concentrations are expected to be achieved within two days of dosing. Quetiapine is unlikely to interfere with the metabolism of drugs metabolized by cytochrome P450 enzymes. Some differences in the pharmacokinetics of quetiapine were noted between children/adolescents (10-17 years of age) and adults. When adjusted for weight, the AUC and Cmax of quetiapine were 41% and 39% lower, respectively, in children and adolescents compared to adults.
When pregnant rats and rabbits were exposed to quetiapine during organogenesis, there was no teratogenic effect in fetuses. Doses were 25, 50 and 200 mg/kg in rats and 25, 50 and 100 mg/kg in rabbits which are approximately 0.3, 0.6 and 2-times (rats) and 0.6, 1 and 2-times (rabbits) the MRHD for schizophrenia of 800 mg/day based on mg/m2 body surface area. Maternal toxicity (observed as decreased body weights and/or death) occurred at 2 times the MRHD in rats and approximately 1-2 times the MRHD (all doses tested) in rabbits.
Bipolar Disorder
Do not stop using Seroquel suddenly, your condition may get worse, or you could have unpleasant symptoms (such as trouble sleeping, nausea and vomiting). Drinking alcohol while taking quetiapine could increase your risk of sleepiness, dizziness, feeling lightheaded, or fainting upon standing. Because quetiapine can cause your body temperature to increase, try to avoid becoming too hot or dehydrated.
Dosage for bipolar disorder (depressive episodes)
High blood sugar can occur in people with or without diabetes. Symptoms can include feeling very thirsty or hungry, needing to urinate more than usual, feeling weak or tired, or having fruity-smelling breath. If your doctor has directed you to use this medication, your doctor or pharmacist may already be aware of any possible drug interactions and may be monitoring you for them.
Post-Traumatic Stress Disorder (PTSD)
The use of drinks that contain alcohol raises your risk of this side effect. If you drink alcohol, talk to your doctor about whether this drug is safe for you. Don’t take this drug again if you’ve ever had an allergic reaction to it. Examples of drugs that can cause interactions with quetiapine are listed below. Quetiapine is available as the brand-name drugs Seroquel (immediate-release tablet) and Seroquel XR (extended-release tablet).
If you are breastfeeding, consult your healthcare provider to determine if Quetiapine is the best option for you. They may recommend monitoring your baby for any signs of side effects or suggest alternative treatments. Take Quetiapine exactly as prescribed by your healthcare provider.
Does quetiapine interact with other medicines (drug interactions)?
If hypotension occurs during titration to the target dose, a return to the previous dose in the titration schedule is appropriate. Table 4 shows the percentage of adult patients with changes in total cholesterol, triglycerides, LDL-cholesterol, and HDL-cholesterol from baseline by indication in clinical trials with SEROQUEL. The mean change in fasting glucose from baseline was 3.2 mg/dL and mean change in 2-hour glucose from baseline was -1.8 mg/dL for quetiapine. There are no systematically collected data to specifically address switching patients with schizophrenia from antipsychotics to SEROQUEL, or concerning concomitant administration with antipsychotics. While immediate discontinuation of the previous antipsychotic treatment may be acceptable for some patients with schizophrenia, more gradual discontinuation may be most appropriate for others. In all cases, the period of overlapping antipsychotic administration should be minimized.
Medications like Buspirone (Buspar) or SSRIs such as Fluoxetine (Prozac) can be alternatives. Cognitive behavioral therapy (CBT) and relaxation techniques may also help alleviate symptoms. Mood stabilizers like Lithium or Valproate (Depakote) are commonly used.
Taking Seroquel may cause eye problems, such as cataracts or glaucoma. If you already have one of these conditions, the drug may worsen them. For this reason, your doctor may recommend that you get an eye exam before starting Seroquel. And during your Seroquel treatment, be sure to tell your doctor about any changes in your vision. Seroquel can increase your blood sugar level, which can lead to diabetes.
Tell your healthcare provider if you have any of the following signs of low thyroid hormones. Some people who have taken quetiapine have reported having seizures. There are many different types of seizures and some of them are not easy to recognize.
